Instructions for Using the UV-Vis Spectrophotometer

 

Preparation of Sample:

 

The UV-Vis spectrophotometer is a sensitive instrument.  Your sample must be dilute to obtain a proper spectrum.  If your sample is a liquid, place one drop of your sample in a small test tube, and fill it full with HPLC grade methanol.  If your sample is a solid, place 1-2 crystals in a small test tube, and fill it full with HPLC grade methanol.

 

Obtaining a Spectrum on the Genesys 10:

 

1.) Turn ON switch on back of instrument and wait for warm up and calibration self/check procedure. When ³Scanning² menu comes on screen‹(Test name UV scan<saved>)

Press green button below ³RUN TEST² at bottom of screen

 

2.) Obtain a quartz cuvette from your T.A.  Fill the quartz cuvette full with HPLC grade methanol, and place the sample cuvette in the B position of sample turret. Make sure the cuvette is clean. (Inspect for fingerprints and filth.)

 

3.) Press green button below ³Collect Baseline² 

 

4.) After baseline is collected, remove reference/blank sample and insert your sample in position 1 in sample turret.  Press 1 on the turret diagram to move turret in position to read sample 1 

 

5.) Press green button below ³Measure Sample²

 

 

If no new baseline is needed, load next sample in turret 1 position and Press ³Measure Sample²

 

If new baseline is needed, insert reference/blank in B position of turret, press B on turret diagram to move turret to B position and press button below ³Collect Basline²

 

 

Each run will print automatically when sample run is completed.

 

 

Generally we only need to collect data from about 215 ­ 400 nm since our unknowns are not highly colored in the visible region of the spectrum.
